چکیده

The distribution of RNase activity and the occurrence of hyperbasophilic foci and tumors have been examined in rat liver during diethylnitrosamine carcinogenesis. A focal loss of RNase activity was found to occur in the hyperplastic liver nodules prior to the development of hy perbasophilic foci and hepatomas. The RNase-negative areas were widely distributed in livers of rats treated with diethylnitrosamine and so were the hyperbasophilic foci and the tumors. The results indicate that temporal and topographic correlations exist among these phenomena and suggest that a deficiency of RNase activity may play a role in the development of hyperbasophilic foci and hepatomas.

Histochemical activity of alkaline and acid nucleases in the rat liver parenchyma during N-nitrosomorpholine carcinogenesis.

SUMMARY @ The activity of alkaline and acid DNase and RNase was histochemically investigated in the rat liver during different stages of N-nitrosomorpholine-induced carcinogenesis. The activity of acid and alkaline nucleases was considerably decreased in focal areas and later in the hyperplastic nodules, whereas the surrounding liver parenchyma demonstrated normal activity of these enzymes. Nuc...

Histochemical studies on rat liver proteins during 4-dimethylaminoazobenzene carcinogenesis.

Areas of preneoplastic livers characterized by intense RNA staining apparently represent the sites of neoplastic transformation. These hyperbasophilic foci, as well as the hepatomas, showed a marked decrease in the stain ing intensity of the cytoplasmic proteins in sections stained by the mercury-bromophenol blue method or the MillÃ3nreaction. Following extraction of the RNA, how ever, protein ...

MicroRNAs involved in neoplastic transformation of liver cancer stem cells

BACKGROUND The existence of cancer stem cells in hepatocellular carcinoma (HCC) has been verified by characterizing side population (SP) cells based on efflux of Hoechst 33342 dye from stem cells. Recent advances in microRNA (miRNA) biology have revealed that miRNAs play an important role in embryonic development and tumorigenesis. However, it is still unclear which miRNAs participate in the ne...

Amplification, rearrangements, and enhanced expression of c-myc in chemically induced rat liver tumors in vivo and in vitro.

c-myc expression, amplification, and rearrangement were studied in neoplastic hepatic nodules of rats after diethylnitrosamine treatment, in hepatocellular carcinomas induced by N-methyl-N-nitrosourea, and in tumorigenic liver cell lines derived from rat hepatocytes transformed by diethylnitrosamine in vivo. Steady-state levels of c-myc transcripts, as measured by Northern blot hybridization, w...

Production of monoclonal antibodies to preneoplastic liver cell populations induced by chemical carcinogens in rats and to transplantable Morris hepatomas.

Monoclonal antibodies (moabs) to neoplastic and preneoplastic liver cells in rats have been selected to follow cellular changes in the livers during chemical carcinogenesis. The moabs were induced by immunizations of BALB/c mice with four partially purified liver cell preparations: 1) oval cells induced in male Fischer rats fed 0.05% N-2-acetylaminofluorene in a choline deficient diet: 2) prene...
